Biotin was covalently attached to antibodies, antigens and enzymes, and the effects of this labeling on the antigen and antibody binding capacity and on enzymatic activity were tested. Based on avidin-biotin interaction, the labeled proteins were used in quantitative enzyme-immunoassay and emical staining procedures. Two procedures were developed.
Techniques harnessing the strong interactions of the avidin–biotin system are frequently used in biosensing and protein separation. Hydroxyapatite (Ca10(PO4)6(OH)2, HAp) is a calcium phosphate compound known to have a strong affinity for various proteins. This technique involves performance of an indirect immunoperoxidase sandwich (including development of the enzyme reaction) followed by an unlabelled immuno-alkaline phosphatase sandwich (the APAAP method).

As in all immunologic techniques using labeled compounds, the antibodies employed in immunoenzymatic procedures can be distinguished as unlabeled ones, . those that take part in the first reaction with the antigen, and as enzyme-labeled antibodies that are added secondarily and, usually, in high excess.
